A model of the three-dimensional structure of Bcl-2. This model is based on the three-dimensional structure of Bcl-XL (pdb file 1LXL[103]) and the more recent structures of Bcl-2 in which the unstructured loop was excised and replaced by the corresponding sequence from Bcl-XL (pdb file 1GJH[104]). Protein is rendered using colours to represent the similarity between the sequence of the protein and the sequences of peptides selected for affinity to Taxolâ„¢. Red indicates a high level of similarity, blue a very low level. The highest level of similarity is in the flexible 50 amino acid loop, which has no structure detectable either from X-ray crystallography or nuclear magnetic resonance spectroscopy, and is thus highly mobile in solution
